Research has observed that NAD+ availability tends to fall with age and metabolic stress, which has made restoring NAD+ a major focus of healthy-ageing science.

What is nicotinamide riboside (NR)?

Nicotinamide riboside is a form of vitamin B3 that the body can convert into NAD+. Because it is a direct NAD+ precursor, it is widely sold as a longevity and cellular-energy supplement and is actively being studied in humans.

What does the current research show?

A completed clinical study at Cambridge University Hospitals NHS Foundation Trust — NCT03432871, "The Role of Nicotinamide Riboside in Mitochondrial Biogenesis" — examined how NR is taken up by the body (its bioavailability) and its relationship to mitochondrial energy production. It was a small study (13 participants), which is typical of this early stage of research.

This and similar trials investigate whether NR raises NAD+ and supports mitochondrial function — they do not prove that NR slows ageing or treats any disease. Most NAD+ research to date is small and exploratory. Look for larger, peer-reviewed trials before drawing firm conclusions.

How to think about NAD+ supplements

  • Promising, not proven. NAD+ precursors are a genuine area of science, but human evidence for longevity benefits is still limited.
  • Quality and dose matter. Look for products that name the exact form (e.g. nicotinamide riboside) and dose, ideally third-party tested.
  • Foundations first. Sleep, exercise and a good diet have far stronger evidence for healthy ageing than any single supplement.
